Narodila se v roce 1990 v Třinci. Studovala Janáčkovu konzervatoř v Ostravě, po jejímž absolvování nastoupila do Těšínského divadla. V průběhu těšínského angažmá hostovala i v Národním divadle moravskoslezském v Ostravě, v letech 2014–2019 pak byla v angažmá ve Východočeském divadle v Pardubicích, kde ztvárnila řadu velkých rolí – jmenujme alespoň Violu v Zamilovaném Shakespearovi, Marii v Konci masopustu nebo Karlu v Kalibově zločinu.
Od sezóny 2019/2020 je členkou činoherního souboru DJKT. Zde se již stihla zhostit několika výjimečných rolí. Nejdříve zaujala komediální rolí paní Sirrelliové v představení Každý má svou pravdu. Aby zanedlouho přímo zazářila studií postavy Beccy v rodinném dramatu Králičí nora. Odlišnou polohou půvabné a život milující Maryšky v Postřižinách prokázala Jana Ondrušková široký rejstřík svého hereckého umění. Děti ji zase mohly ocenit v nastudování Kukačky, Čmeláka, Vodénky a Panenky v pohádce O Křemílkovi a Vochomůrkovi.
Jana Ondrušková miluje neotřelý humor, přírodu a dobře vychlazené pivo.
